2X General Serum Diluent is formulated for diluting goat, rabbit, mouse, avian, and human sera, as well as mouse ascites fluid and cell culture media, for testing in sandwich ELISA configurations. It can also be used for dilution of many types of serum samples in antigen-down ELISA formats. Our 2X concentrated version is helpful for shipping purposes and produces less waste. - General Serum Diluent is formulated to provide a stable, protein-friendly environment for dilution of serum, cell culture, ascites, urine, and other aqueous-based biological samples for evaluation within an ELISA format. It can serve as an excellent generic matrix in which to dilute antigen standards when preparing standard curves within antibody-sandwich formats. It also provides an excellent serum dilution medium for antigen down ELISAs in the assessment of a humoral immune response to a particular disease state or immunization regimen., , General Serum Diluent provides a mammalian protein-buffered, neutral pH environment that is highly compatible with antibody-antigen interactions. Inclusion of a protein component helps minimize the degree of non-specific IgG adsorption onto coated and blocked ELISA plate wells during antigen-down screening of serum or plasma samples., , Bulk volumes and custom packaging are available upon request. - 1. Dilute the standard curve, controls, and the samples as necessary. ICT offers several formulations of sample diluents in which to prepare the samples., 2. Prepare 1X working strength assay diluent by diluting 1:2 in diH2O. Add 1 part General Assay Diluent, 2X to 1 part diH2O. Mix well., 3. Pipette 50-100 μL General Assay Diluent per well into every well of the plate., 4. Pipette 50-200 μL of each standard, control, and sample into the plate., 5. Run the assay according to the specific ELISA protocol., 6. Analyze the data. Because all of the wells, including the standards and controls, received the same volume of assay diluent, there is no need to account for this dilution when calculating the results.
